Tower Hamlets Council selected the London contractor as preferred bidder following a competitive flexible procedure, which included Morgan Sindall and Chalegrove Properties also bidding the job.
The job will see the existing leisure centre in Shadwell demolished and replaced with a modern facility alongside 30 new homes.
Guildmore will also take the design through RIBA Stage 4 with the opportunity to deliver associated landscaping and public realm works.
The contract will be entered in May, with completion targeted for April 2029.
A standstill period runs until 17 April, with contract signing pencilled in for 27 April.
The project forms part of Tower Hamlets’ wider drive to upgrade community leisure provision while unlocking small-scale housing delivery on public land.
